教室の概要

人間の脳は、神経細胞(ニューロン)とそれを支える膠細胞(グリア)から成り立ち、ニューロンはシナプスを介してネットワークを広げ、情報の伝達を行って います。私たちは、シナプスで働く分子(遺伝子とタンパク質)の機能を研究することにより、脳が働くメカニズムの解明を目指しています。また、これらの機 能が破綻することによって引き起こされる精神・神経疾患の解明を目指して研究しています。

研究テーマ

1. Neurexin/Neuroligin と精神疾患との関係の解明

Neurexin およびNeuroligin は異なるグループに属する細胞接着因子のファミリーで、Neurexin はシナプス前終末、Neuroligin はシナプス後終末にそれぞれ局在し、カルシウム依存的に両者が結合することにより、シナプス認識の特異性および機能獲得に寄与していると考えられていま す。近年、NeurexinおよびNeuroligin 、さらにはこれらと細胞内で相互作用する分子の遺伝子異常が自閉症をはじめとする精神疾患の患者から見つかったことから、 Neurexin/Neuroligin によって維持されるシナプス機能の破綻が、これらの疾患の病態と関係しているのではないかと考えられるようになってきました。
私たちは、 Neurexin、Neuroligin の各種アイソフォームを網羅する遺伝子改変マウスを作成し、これらのシナプス機能を生化学的、薬理学的、電気生理学的、形態学的手法を用いて解析すると共 に精神疾患関連行動の解析をし、これらの因果関係を調べています。さらに、これらを精神疾患モデルマウスとして評価・確立することを目指して研究を行って います。

2. γ-セクレターゼの機能異常と神経変性疾患との関係の解明

Nicastrinはγ-セクレターゼ複合体を構成する一回膜貫通タンパク質です。これまで私たちは、Nicastrin がγ-セクレターゼの基質の認識および酵素活性に必須であり、この欠損細胞ではγ-セクレターゼの機能が完全に失活することを見出してきました。また、 Nicastrin が大脳皮質および海馬の興奮性シナプスで欠損したマウスでは神経変性が起こることも見出しました。現在、Nicastrin欠損マウスのシナプス機能を解 析すると同時に、γ-セクレターゼの異常が神経変性を起こすメカニズムについて研究を行っています。
