    好塩基球は最近Th2応答の開始に重要な細胞として注目を集めていますが、好塩基球のTh2誘導サイトカインIL-4の産生に至る細胞内シグナル伝達経路はよく分かっていません。この研究では、これまで様々なimmuoreceptorのシグナルに必須であることが知られていたFcRγが、IL-3受容体βc鎖にも結合しており、好塩基球にIL-4産生を誘導するIL-3のシグナル伝達に必須の役割を果たしていることを明らかにしました。そして、ITAMを持つアダプター分子がサイトカイン受容体のシグナルに関与しているという新たな知見を提供しています。

    IL-15はNK細胞の分化、増殖に必須のサイトカインとして良く知られていますが、この研究ではIL-15がNK細胞にインターフェロンγの産生を誘導してプレB細胞の増殖を抑制することを明らかにしました。さらに、IL-15によって増殖するNK細胞はB220を発現しており、活性化NK細胞として最近報告された細胞に対応すると考えられました。IL-15は炎症時に産生されますので、炎症時に見られる骨髄でのプレB細胞抑制に活性化NK細胞が関与している可能性が示唆されます。

    ヒト乾癬などに代表される炎症性皮膚疾患の多くは多因子疾患であり、複数の遺伝子によってその発症が規定されています。我々が以前に報告したIRF-2欠損マウスに自然発症する皮膚炎は背景となるマウス系統によってその重症度が異なり、今回のリンケージ解析の結果、複数の遺伝子が発症に関係している事が明らかになりました。さらにこの遺伝的制御機構が IFN-α/βシグナルとその下流の皮膚IFN-γ発現に向けられていることも明らかになり、ヒト皮膚炎症発症の分子機構の理解につながることが期待されます。

    好塩基球は、これまで主にアレルギーの効果相で機能すると考えられてきましたが、その数がもしも適切に制御されなければアレルギー反応につながるTh2分化過剰を招き得ることが明らかになりました。転写因子IRF-2は、好塩基球の分化やIL-4産生には影響を与えないけれども、その末梢でのIL-3に対する増殖応答をネガティブに制御して、生体のTh1/Th2バランスの維持に関与しています。
